Blue et al. (2025)  Multi-ancestry meta-analysis identifies genetic modifiers of age-at-onset of Alzheimer’s disease at known and novel loci

We performed a genome-wide association study for age-at-onset of Alzheimer’s disease (AD) in two diverse cohorts. The 16 loci significantly associated with age-at-onset of AD in the meta-analysis include several AD risk loci as well as novel loci. Together, the genes implicated represent several AD biological domains, including structural stabilization, immune response, and the synapse. Secondary analyses revealed APOE adjustment influenced effect size estimates for variants across the genome, some with the same or greater absolute value as well-established risk alleles.
